Studies on Paeoniflorin

Paeoniflorin is a white fine powder chemical compound with Molecular Formula:C23H28O11 and Molecular Weight: 480.47 . It is one of the major constituents of a herbal medicine derived from Paeonia lactiflora. Further study demonstrated that chronic treatment with Paeoniflorin suppressed the activations of JNK and p38 MAPK, but enhanced ERK activation. And PF could reverse ischemia-induced activation of NF-κB signaling pathway. Moreover, our in vitro study revealed that PF treatment protected against TNFα-induced cell apoptosis and neuronal loss.

Taken together, the present study demonstrates that Paeoniflorin produces a delayed protection in the ischemia-injured rats via inhibiting MAPKs/NF-κB mediated peripheral and cerebral inflammatory response. Our study reveals that PF might be a potential neuroprotective agent for stroke.

We demonstrated that Paeoniflorin is an effective compound in the modulation of autophagic activity. Treatment with MPP+ significantly inhibited autophagy and induced apoptosis in PC12 cells. However, co-treatment with Paeoniflorin upregulated the autophagy and clearly showed neuroprotective effects. 

In addition, the CMA pathway was activated as a compensatory mechanism, while Paeoniflorin efficiently maintained the CMA activity near the normal range. We also exposed PC12 cells to an acidic medium, since acidosis is a common cause of mitochondrial dysfunction and neuron degeneration. 

Similarly, the deficiency of autophagy and the overactivity of CMA were both attenuated by Paeoniflorin. Thus, the key finding of the present study was that Paeoniflorin potently protected PC12 cells against MPP+ or acidosis-induced injury by upregulating the autophagic pathway.

In summary, the main findings of this study are: (1) Paeoniflorin and Ami both potently protected PC12 cells against MPP+ or acidosis induced injury by upregulating the autophagic pathway; (2) Paeoniflorin reduced calcium in flux by inhibiting ASIC1a channels; (3) the activity of ASICs altered the function of ALP.

Permethrin is a common synthetic chemical, widely used as an insecticide, acaricide, and insect repellent. Nevirapine can kills HIV virus


Nevirapine is a white or off-white crystalline powder, also marketed under the trade name Viramune. It is a non-nucleoside reverse transcriptase inhibitor used to treat HIV-1 infection and AIDS. Longer nevirapine prophylaxis to prevent postnatal HIV infection appears to benefit children, but the gains seem to disappear unless they are treated during the entire breastfeeding period, researchers said here at the Conference on Retroviruses and Opportunistic Infections.

Study on the effects of (-)-Epigallocatechin gallat on Cancer

(-)-Epigallocatechin gallat is also known as epigallocatechin-3-gallate, is the ester of epigallocatechin and gallic acid, and is a type of catechin, we also call it EGCG. Green tea is a popular drink consumed daily by millions of people around the world. Previous studies have shown that some polyphenol compounds from green tea possess anticancer activities. However, systemic evaluation was limited. We determined the cancer chemopreventive potentials of 10 representative polyphenols and explored their structure-activity relationship. 

The effect of the 10 polyphenol compounds on the proliferation of HCT-116 and SW-480 human colorectal cancer cells was evaluated using an MTS assay. Cell cycle distribution and apoptotic effects were analyzed by flow cytometry after staining with propidium iodide (PI)/RNase or annexin V/PI. Among the 10 polyphenols, EGCG showed the most potent antiproliferative effects, and significantly induced cell cycle arrest in the G1 phase and cell apoptosis. 

When the relationship between chemical structure and anticancer activity was examined, C and EC did not show antiproliferative effects, and GA showed some antiproliferative effects. When C and EC esterified with GA to produce CG and ECG, the antiproliferative effects were increased significantly. 

A similar relationship was found between EGC and (-)-Epigallocatechin gallat. The (-)-Epigallocatechin gallat group significantly enhanced catechin's anticancer potential. This property could be utilized in future semi-synthesis of flavonoid derivatives to develop novel anticancer agents.
